This course will focus on the chemical synthesis and purification of bio-orthogonal functional molecules and their application in bio-orthogonal reactions. We will present chemical methods for molecule preparation and perform various bio-orthogonal reactions with the derivatives. Additionally, we will investigate the advantages and limits of these reaction types, as well as explore the many analytical techniques we have to offer e.g. NMR and mass spectrometry.

Correlative Light- and Electron Microscopy (CLEM), Electron Tomography and Advanced Sample Preparation Techniques

This course will cover advanced electron microscopy (EM) techniques. We will perform sample preparation with cryo-immobilization to get access to near-to-native EM samples (i.e. high pressure freezing and freeze substitution). We will also discuss classical sample preparation techniques. Furthermore, we will acquire own images at transmission and scanning electron microscopes in order to learn electron tomography and correlative light- and electron microscopy. Finally, we will learn how to use software packages for EM image processing and analysis.

This workshop focuses on the introduction of methods and tools to study the role of lipids and lipid modifying enzymes cellular membranes.

The three day practical lab training will cover the following methods:

1)    Measurement of neutral and acid sphingomyelinase activities in unmodified cells or cells overexpressing or depleted for the respective enzyme.

2)    Visualization of incorporation and trafficking of lipids by fluorescence microscopy (fluorescent lipid analogues, functionalized sphingolipids (living cells), ceramide-specific antibodies (fixed cells) in cell types (of choice).

3)    Analysis of sphingomyelinase inhibitor treated or lipid loaded cells:

        a) quantification of membrane order by flow cytometry using fluorescent dye di-4-ANEPPDHQ;

        b) determination of cytoskeletal activity by flow cytometry and microscopy analysis.

In this workshop we will discuss key aspects of how leukocytes mediate immunity.

These will include the specificity of immune responses for certain pathogens as well as leukocyte migration as an essential mechanism for generating and maintaining immunity. 

Hands-on work will focus on the use of flow cytometry as one of the central techniques for studying immune cell biology.

In this workshop we will discuss key aspects of molecular pathomechanisms of bacterial, viral and fungal infectious diseases in humans.

Students will learn the basics of medical microbiology and virology, i.e. microscopy, sample preparation, culture methods and laboratory diagnosis.

Hands-on work will focus on classical and molecular diagnostic assays as well as techniques used in cellular microbiology, e.g. infection assays.
