Cancer presents an eminent burden of disease worldwide and a pressing need for innovative therapeutic strategies. Deregulated transcription is a hallmark of oncogenic transformation. We study how the transcription machinery is composed and regulated in cancer cells. While ubiquitylation and degradation of Pol2 is functionally important to remove arrested Pol2 from damaged DNA, the role of ubiquitylation in transcription in unstressed cells is stillunderexplored. Here, we will investigate the impact and mechanistic details of Pol2 ubiquitylation in mammalian cells.
